How & why

William Howard Stein, from a New York Jewish family, worked at the Rockefeller Institute, where with Stanford Moore he developed automated methods to separate and measure amino acids. Using these tools they determined the complete sequence and active structure of ribonuclease, one of the first enzymes to be fully mapped, linking its chemical structure to how it works. This achievement brought them a share of the 1972 Nobel Prize in Chemistry.
