Horwich, a biologist and physician at Yale, discovered in 1989 that proteins do not always fold unaided: he identified the chaperonins, barrel-shaped molecular machines that provide a protected chamber in which newly made proteins fold correctly. This overturned the textbook view of protein folding and illuminated diseases of misfolding, earning him the Lasker, Shaw and Breakthrough prizes.
